Jim:
The most straightforward way is to use Partition Magic or a similar
program. However, there are a few other considerations:
1. Depending on your hardware, your motherboard may not be able of
handling a partition larger than a certain size - where "certain"
depends on your operating system and the age of your motherboard.
2. There is an interrelationship between partition size and space usage.
3. There is a school of thought which says that multiple partitions are
a good thing. The argument is that having the OS and applications on one
partition, the data on another, and backups and archives on a third
partition not only simplifies system administration, but it makes
backing up much simpler, and can also reduce the impact of a hardware
failure.

Not knowing what operating system and hardware you have, nor your
reasons for wanting to put everything in one partition, that's about the
best that I can do.
