There's a program called "Talking Email"

As described at http://4developers.com/talkmail/index.htm

You can specify if you want to hear who sent the message, its subject, and
how many lines of the message should be read, and be notified for mail from
a specific individual. It also gives a choice if sounds.

I haven't tried it, and don't know how it interacts with screenreaders...
but it's free...

(Well, if you want different animated characters on the screen then you
gotta pay).
