Some Android devices, Samsung and a few LG models, have physical home 
buttons, which you can triple-click to toggle accessibility, but this 
feature needs to be enabled in Accessibility settings before it can be 
used. As far as first-time setup, I think the triple-click home doesn't 
work because the accessibility shortcut (which may be called direct 
access) hasn't been enabled. I know it didn't on older devices, but I 
don't remember for newer ones.

On most Android devices, you start accessibility during set-up by 
placing two fingers on the screen (about an inch apart) and holding them 
there until the device says, "TalkBack on."

You can also restart TalkBack from off at any time after you enable the 
Accessibility Shortcut (Go into Settings > Accessibility > Accessibility 
Shortcut, tap On, then OK). To restart TalkBack, wake the device up (if 
it isn't already), hold down the power button for about 3 seconds (until 
you feel a vibration on your phone or hear the default notification tone 
on your tablet), then hold two fingers on the screen until you hear, 
"TalkBack on."

If all you want to do is suspend or resume TalkBack (e.g., so you can 
use a self-voicing app), however, just hold down the volume-up and 
volume-down keys at the same time. I believe the feature is enabled by 
default, but it doesn't hurt to double-check in settings > Accessibility 
 > TalkBack > Settings.
