Tweakui has been known to cause this problem by hiding drive letters.
See if it's installed and adjust accordingly.

I have a Kingston Datatraveler encrypted flash drive. It works fine on
most computers. Plug it in and the Privacy Vault screen pops up
(sometimes...sometimes you need to run the executable, but that's not
really the problem), you enter your password and you're good to go. What
you see then is the Kingston device and the next lettered drive with the
files on the flash drive.
 
In one computer when the password is entered, it says it's looking for a
specific letter drive, but it can't find it and you should insert the
disc/drive. I think it's that next lettered drive it's looking for, but
since the flash drive is already in the USB port, there's nothing more
to insert.
 
Thoughts or ideas to get this one computer to recognize the drive? It's
not just me. Several people with the same drive are having similar
problems.
