almost sounds like you are trying to transmit out of band or something.
Do you use memory channels or the VFO when trying to use 2m?
Where are you trying to transmit?
You may have inadvertently set the radio to transmit on a frequency out of 
the tx band of the radio.
Easiest way to check is to press the rev/split button, which is the round 
one to the right of the main VFO dial.  This will emit a high beep, then 
queery the frequency with whatever button you are using to get frequency.
You may have somehow gotten the tx frequency way off especially if you are 
using VFO mode instead of MR mode.
If you are using MR mode just turn the radio off and back on again and 
select that channel to reset all parameters.
BTW if you get a low beep when you press that round rev/split button, it 
means you are set to simplex mode with no shift.
The zero key will toggle plus, minus and simplex.
You didn't turn tx inhibit on because that works globally in the radio and 
not by band.
